Panthenol — pro-vitamin B5 — is one of the few “hair vitamins” with data on the hair fiber itself, although all of it is laboratory data. In one study on hair fibers, deuterium-labeled panthenol was found to penetrate the protein regions of the hair cortex, and hair exposed to panthenol had a higher break stress and elastic modulus than control hair; most of the authors work for a cosmetics company and the abstract does not state the concentration. Panthenol is also slightly hygroscopic, and the CIR Expert Panel lists hair conditioning among its functions. The sources we checked contain no consumer trial of a panthenol hair spray, so the B5 hair spray Butter-C develops promises what can be measured on the finished product: soft, easy-to-comb hair, moisture retained in the hair fiber, and reduced breakage during combing where measured — not restoration of damaged hair or thicker hair strands. The technical work lies in a clear water-based solution thin enough to mist: choosing D- or DL-panthenol, keeping cationic conditioners within the limit for leave-on hair products, and holding a pH that suits panthenol while hair stays non-greasy after several hours. Formula base
Panthenol, cationic conditioners and a water base
A B5 hair spray is a very thin water-based solution; every ingredient must dissolve completely so the solution stays clear and mists evenly.
D-panthenol or DL-panthenol
Same INCI name, Panthenol. D-panthenol (dexpanthenol, CAS 81-13-0) is a viscous, slightly hygroscopic liquid that may partly crystallize on standing; DL-panthenol (CAS 16485-10-2) is a mixture of both isomers. Butter-C’s stock material is D-panthenol; the certificate of analysis must state the form — the same choice as on the B5 gel page.
Panthenol level
The 1% and 5% human data are skin data (reduced water loss through the skin), not hair data; the hair-fiber study above does not report a concentration in its abstract. The level in a spray is chosen by feel on hair and stickiness (formulation experience).
Cationic conditioners alongside
Polyquaternium-10 at a low level for detangling, Cetrimonium Chloride to reduce static. Cetrimonium Chloride in leave-on hair products is limited to 1.0% (individually or as a sum with steartrimonium chloride, for purposes other than preservation — ASEAN Annex III, Ref 286; as a preservative, Annex VI, Ref 44 applies, maximum 0.1%); the commercial material is a water solution, so it must be converted to active content using the certificate of analysis.
Water base, humectant and fragrance
Purified water, a moderate level of Glycerin, a water-soluble silicone (PEG-12 Dimethicone) for light slip; fragrance solubilized with PEG-40 Hydrogenated Castor Oil to keep the solution clear, or no fragrance.
Key technical points
Four things specific to a B5 hair spray
Panthenol mixes easily into water, but a hair spray still has to be clear, mist finely, stay non-greasy and keep its content through shelf life.
A clear solution thin enough to spray
Panthenol is freely soluble in water; what raises viscosity is the cationic polymer — kept low so the mist is fine rather than droplets. Clarity is checked at room temperature and after cold storage.
pH and heat when adding panthenol
PubChem notes that dexpanthenol is more stable than pantothenate salts at a pH of about 3–5, and that prolonged heating causes racemization. Panthenol is added in the cool-down phase; the finished pH is set by assaying panthenol across the stability time points, not by habit.
Keep cationic and anionic ingredients apart
Cetrimonium Chloride and Polyquaternium-10 carry a positive charge; anionic thickeners such as Carbomer and Xanthan Gum can cause haze, precipitation or weaker conditioning. Clarity and viscosity are tested on the full formula.
Light, non-greasy and safe to spray
A spray may land on the scalp, forehead and nape several times a day: conditioners stay low and hair is assessed after several hours. Mist can reach the eyes and nose: the label directs users to spray onto hair, away from the face, avoiding the eyes (good practice). Concentrated Cetrimonium Chloride is corrosive to skin and eyes according to the SCCS — the weighing area requires goggles and gloves.

Everyday moisture B5 spray
Panthenol, Glycerin and a low level of Polyquaternium-10; fragrance-free or lightly scented; for hair dried by sun and air conditioning, used on damp or dry hair.
Detangling B5 spray
Panthenol with Cetrimonium Chloride and PEG-12 Dimethicone; detangles wet hair after washing and reduces static when combing — performance measured by combing force on hair tresses.
B5 – rice protein spray for permed or colored hair
Panthenol combined with hydrolyzed rice protein; a Vietnamese rice story; claims stop at soft, easy to comb and moisture retained in the hair fiber.
Packaging is designed with the formula
Fine-mist spray bottles; the actuator determines how fine the mist is. Check for nozzle clogging when panthenol and polymers dry at the tip (formulation experience).

What our R&D lab measures on every sample
Below are the parameters Butter-C tracks during formula development and scale-up. Specific limits are agreed with you on the approved sample and then written into the product’s in-house specification.

- Checked at room temperature and after cold storage; no haze, film or precipitate.

- Measured at 25°C on every lot; kept within the range agreed for panthenol stability and the preservative system.
- Viscosity and spray pattern
- Checked on the actual actuator to be used: mist fineness, evenness and dose per actuation.
- Incoming panthenol form
- Certificate of analysis checked for every lot: D or DL form, content.
- Panthenol content
- Assayed in the finished product and across the stability time points when the label states a B5 percentage.
- Cetrimonium Chloride as active
- Converted from the raw-material percentage using the certificate of analysis; the sum with steartrimonium chloride stays within 1.0% for leave-on hair products.
- Performance on hair tresses
- Wet and dry combing force, greasiness after several hours; tensile break force or breakage during combing if you want to use the matching claim.
- Microbiology and preservative efficacy test
- Tested at a GLP / ISO 17025-accredited laboratory; preservative efficacy (challenge) test to ISO 11930.
- Irritation and sensitization
- Volunteer testing before the formula is finalized; contact allergy to panthenol is uncommon but has been reported.
- Packaging compatibility
- Monitored for nozzle clogging, leakage and discoloration of the solution in the bottle.
Claims
Claims that match the level of evidence
Panthenol hair data are laboratory measurements on hair fibers, mostly from authors working for companies. The often-quoted 2–5 µm increase in hair fiber diameter comes from a five-ingredient leave-on formula (caffeine, niacinamide, panthenol, dimethicone and an acrylate polymer), so panthenol’s share cannot be separated — and “thickens hair strands” is among the claims Appendix 03-MP (Circular 06/2011/TT-BYT) does not accept. Butter-C reviews the wording on labels and marketing materials while preparing the notification dossier.
Leaves hair soft and easy to comb
Supported by combing-force measurements on hair tresses. Instead of: restores damaged hair, rebuilds hair structure.
Helps the hair fiber retain moisture (when measured on the finished product)
Instead of: deep hydration for hair, restores dry, damaged hair.
Helps reduce breakage during combing (when measured on the finished product)

Contains pro-vitamin B5 (panthenol)
State the content when it can be assayed. Instead of: vitamin that nourishes the follicle, pure B5.

When the formula contains a cationic conditioner and has been assessed on hair tresses. Instead of: permanently smooth hair.
Directions and precautions
Spray onto hair, away from the face, avoiding the eyes; test first on a small area of skin and stop use if the scalp becomes irritated. Instead of: hypoallergenic, safe for all skin types.
